技术原理:
酶联免疫吸附试验(ELISA)是一种实验室常用的检测方法, 广泛应用于测量溶液中的分析物(通常是抗体或抗原)的浓度。与其他基于抗体的检测方法不同的是, 酶联免疫吸附实验(ELISA)或酶免疫测定(EIA)通过与固相支持物(通常为聚乙烯多孔板)的结合和系列洗涤步骤, 实现特异性和非特异性相互作用的分离, 并可通过有色产物的形成, 定量分析原始样品中分析物的含量。
ELISA实验快速且简单, 可迅速分析大量平行的样品, 在基础研究、临床前研究和临床诊断实践中广泛使用。
ELISA包括直接法ELISA,间接法ELISA,竞争法ELISA,夹心法ELISA。
双抗体夹心法ELISA是最常见的ELISA类型。使用两种特异性抗体将抗原夹在中间,通常称为抗体对。将捕获抗体包被在微孔板上,加入样品,目标蛋白与抗体结合并固定在板上。然后加入偶联的检测抗体,并与靶蛋白上的其它表位结合。加入底物并产生与样品中目标分析物量成比例的信号。双抗体夹心法ELISA需要两种抗体才能与目标蛋白结合,具有高度特异性。
优点:
特异性高,使用的捕获抗体和检测抗体都是检测物特异性的;
灵敏度高,酶标二抗放大检测信号;
适用于复杂样品, 抗原不需进行纯化即可用于检测;
缺点:
适用于检测分子中具有至少两个抗原决定簇的多价抗原;
不能用于小分子半抗原的检测;
